Analysis of spironolactone polymorphs in active pharmaceutical ingredients and their effect on tablet dissolution profiles BJPS
Resende,Renata Cunha de; Viana,Olímpia Maria Martins Santos; Freitas,Jennifer Tavares Jacon; Bonfilio,Rudy; Ruela,André Luís Morais; Araújo,Magali Benjamim de.
ABSTRACT Spironolactone (SPR) is a steroidal drug administered as a potassium-sparing diuretic for high blood pressure treatment. The drug shows incomplete gastrointestinal absorption due to its poor aqueous solubility. The physicochemical properties of SPR in crystal forms I and II suggest that differences in their aqueous solubility may lead to a lack of bioequivalence between solid-state formulations. In this study, SPR polymorphs in five batches of active pharmaceutical ingredients (APIs) from three manufacturers were characterized using powder X-ray diffraction, infrared spectroscopy, thermal analysis, and solubility measurements. SPR tablets (50 mg) were manufactured in our laboratory using API in pure form II, and API in form II contaminated with...

Analysis of the presence of FLT3 gene mutation and association with prognostic factors in adult and pediatric acute leukemia patients BJPS
Burnatt,Graciele; Licínio,Marley Aparecida; Gaspar,Pâmela Cristina; Ferreira,Arthur Schveitzer; Reis,Manoela Lira; Moraes,Ana Carolina Rabello de; Sincero,Thaís Cristine Marques; Santos-Silva,Maria Cláudia.
ABSTRACT When the FLT3 gene is mutated, it originates a modified receptor with structural changes, which give survival advantage and malignant hematopoietic cell proliferation. Thus, the presence of mutations in this gene is considered an unfavorable prognostic factor. A total of 85 consecutive samples of newly diagnosed untreated patients with AL were included in the study after they provided their informed consent. FLT3 gene mutations were detected by PCR. For the pediatric group, a positive correlation was observed between WBC count and the presence of FLT3-ITD in patients with AML and ALL. Furthermore, children with AML who had the FLT3-ITD mutation showed a tendency to express CD34 in blast cells. In the adult group, the AML patients with FLT3-ITD who...

Animal experimentation and scientific knowledge: a thought style? BJPS
Tréz,Thales de Astrogildo e.
Animal experimentation, besides a research method extensively applied in the production of scientific knowledge, is also considered essential to science and with undeniable historical relevance in advances in human health. In this survey, a questionnaire was applied to a group of researchers involved with research based on non-animal models (n =18), and to another group involved with research based on animal models (n =18). The data analysis was grounded in Ludwik Fleck (1896 -1961) epistemological assumptions. The results suggested that there are at least two thought styles operating in consonance on the same research problem (advances in human health conditions) with significantly different conceptions not only concerning the research practices involved,...
